The expression and clinical significance of Sestrin1 and Sestrin2 in gastric cancer tissues

Abstract:Objective To detect the expression of Sestrin1 and Sestrin2 in gastric cancer tissues and their clinical significance.Methods The clinical medical records of 91 patients with gastric cancer admitted to Guangyuan Central Hospital from March 2019 to September 2020 were collected,and specimens of gastric cancer tissues and corresponding adjacent tissues were collected.The expressions of Sestrin1 and Sestrin2 in gastric cancer tissues and adjacent tissues were detected by immunohistochemical staining,and the relationship between their expressions and the clinicopathological characteristics and prognosis of gastric cancer was analyzed.The survival curve was plotted by the Kaplan-meier method,and the survival differences were tested by Log-rank test.The Cox proportional hazards regression model was used to analyze the factors influencing the prognosis of patients with gastric cancer.Results Compared with adjacent tissues,the positive expression rates of Sestrin1(71.4%vs.30.8%,P<0.001)and Sestrin2(65.9%vs.33.0%,P<0.001)in gastric cancer tissues were significantly increased.The expression of Sestrin1 and Sestrin2 in gastric cancer tissues were related to the depth of gastric cancer invasion,lymph node metastasis and TNM stage(P<0.05).The results of Pearson correlation analysis showed that the positive expression of Sestrin1 was positively correlated with that of Sestrin2(r=0.5134,P<0.05).Compared with those with negative expression of Sestrin1,the median overall survival time of patients with positive expression of Sestrin1(19.5 months vs.28.6 months)was significantly decreased(HR=2.391,95%CI:1.064-4.034,P<0.05);Compared with those with negative expression of Sestrin2,the median overall survival of those with positive expression of Sestrin2(16.9 months vs.29.3 months)was significantly decreased(HR=2.042,95%CI:1.162-3.256,P<0.05).Analysis by the Cox proportional hazards regression model showed that both Sestrin1 expression and Sestrin2 expression were independent factors affecting the prognosis of patients with gastric cancer(P<0.05).Conclusion The expressions of Sestrin1 and Sestrin2 in gastric cancer tissues are significantly increased and can be used as predictive factors for the progression and prognosis of gastric cancer.
