The polymorphism of PSCA gene in cervical squamous cell carcinoma and its clinical significance

Abstract:Objective To investigate the polymorphism of prostate stem cell antigen(PSCA)gene in cervical squamous cell carcinoma(CSCC)and its relationship with high-risk human papillomavirus(HR-HPV)infection.Methods A total of 120 patients with CSCC admitted to Guangyuan Maternal and Child Health Hospital from March 2022 to December 2024 were collected as the research subjects.In addition,120 healthy women who underwent physical examinations during the same period were selected as the control group.Genotyping of the rs2294008 and rs2976392 loci of the PSCA gene was performed by real-time fluorescence quantitative PCR(qPCR).HPV detection and typing were performed using a PCR-based multiplex HPV typing test kit.The expression of PSCA in CSCC tissues was detected by immunohistochemistry.Results Compared with the control group,the frequencies of rs2294008 TT genotype and T allele genotype in the CSCC group decreased(P<0.05).The frequencies of rs2976392 AA genotype and A allele genotype increased(P<0.05).The frequencies of rs2294008 TT genotype and T allele were lower in patients with positive expression of PSCA protein,while the frequencies of rs2976392 AA genotype and A allele were higher(P<0.05).The TT genotype and T allele frequencies of PSCA rs2294008 were lower in patients with HR-HPV infection(P<0.05).The genotype and allele distributions of rs2976392 were not related to HR-HPV infection(P>0.05).Univariate Logistic regression model analysis showed that the genotypes of PSCA rs2294008 and rs2976392 loci were both related to the risk of CSCC occurrence(P<0.05).After adjusting for other confounding factors,the PSCA rs2294008 TT genotype significantly reduced the risk of CSCC(OR=0.032-0.428,P<0.05),and the rs2976392 AA genotype significantly increased the risk of CSCC(OR=2.238-23.276,P<0.05).Conclusion The polymorphisms of PSCA rs2294008 and rs2976392 genes are closely related to the risk of CSCC.The polymorphism of PSCA rs2294008 genes is associated with the risk of HR-HPV infection.Women carrying the rs2294008 T allele are less likely to be infected with HR-HPV.
