Expression and clinical significance of ring finger protein 113A and microRNA-197 in cervical squamous cell carcinoma

Abstract:Objective To investigate the expression and clinical significance of ring finger protein 113A(RNF113A)and microRNA-197(miR-197)in cervical squamous cell carcinoma(CSCC).Methods A total of 104 patients who underwent cervical biopsies at the Department of Gynecology of Xuzhou peace Maternity Hospital from January 2019 to December 2022 were divided into CSCC group(20 cases),high-grade squamous intraepithelial lesions(HSIL)group(20 cases),low-grade squamous intraepithelial lesions(LSIL)group(20 cases),normal cervical tissue HPV-positive group(14 cases),and normal cervical tissue HPV-negative group(30 cases).The expression levels of RNF113A and miR-197 were assessed by quantitative real-time PCR(qPCR),and immunohistochemical staining was used to localize RNF113A in tissue samples.The diagnostic value of RNF113A and miR-197 for CSCC was evaluated using the receiver operating characteristic(ROC)curve.Results Compared with normal cervical tissue HPV-negative group,RNF113A expression was significantly increased in CSCC,HSIL and LSIL groups(P<0.05).As cervical lesions progressed from LSIL to CSCC,miR-197 expression gradually decreased(P<0.05).The results showed that the IHC score of RNF113A in the CSCC group was significantly higher than that in the HPV-positive and HPV-negative normal cervical tissue groups(P<0.05),while there were no statistically significant differences in RNF113A IHC scores between the HSIL,LSIL,and CSCC groups(P>0.05).Additionally,the IHC scores of both the HSIL and LSIL groups were significantly higher than those of the HPV-positive and HPV-negative groups(P<0.05).The Spearman correlation analysis showed that the mRNA expression level of RNF113A was significantly positively correlated with the IHC score(r=0.814,P<0.001).ROC curve analysis showed that the areas under the curve of the relative expression levels of RNF113A and miR-197 in the diagnosis of CSCC were 0.633 and 0.930,respectively.Conclusion The regulation of RNF113A and miR-197 is closely related to the severity of cervical lesions.They have potential as diagnostic biomarkers for early detection and monitoring of the progression of cervical cancer,especially in patients with high-risk HPV infection.
