Expression of activity-dependent neuroprotective protein in pancreatic cancer and its clinical significance

Abstract:Objective To investigate the expression of activity-dependent neuroprotective protein(ADNP)in pancreatic cancer and its clinical significance.Methods The level of ADNP mRNA expression in pancreatic cancer was analyzed using the GEPIA2 database.Immunohistochemicalstaining was used to detect the expression level of ADNP and Ki-67 protein in pancreatic cancer tissues and normal pancreatic tissues,and its relationship with clinical pathological factors and their correlation were analyzed.Cox proportional hazards model was used to analyze the relationship between ADNP protein level and the prognosis of pancreatic cancer patients,and Kaplan-Meier method was used to draw the survival curve.Results GEPIA2 database analysis showed that the level of ADNP mRNA in pancreatic cancer was higher than that in normal adjacent pancreatic tissue(P<0.05).The high expression rates of ADNP and Ki-67 protein in pancreatic cancer tissues were 53.9%(90/167)and 51.5%(86/167),respectively,and 25.1%(42/167)and 0.6%(1/167)in normal adjacent pancreatic tissues,respectively.The expression levels of ADNP and Ki-67 in pancreatic cancer tissues were significantly higher than those in normal pancreatic tissues adjacent to the cancer(P<0.05).The expression of ADNP was correlated with TNM staging,maximum tumor diameter and lymph node metastasis(P<0.05),while the expression of Ki-67 was correlated with TNM staging,maximum tumor diameter,lymph node metastasis,and tumor differentiation degree(P<0.05).ADNP was positively correlated with Ki-67 expression(r=0.256,P<0.05).Kaplan-Meier analysis showed that the survival rate of patients with high expression of ADNP in pancreatic cancer was significantly lower than that of patients with low expression of ADNP(P<0.05).Cox multivariate analysis showed that the expression level of ADNP,TNM stage and the maximum tumor diameter were independent factors affecting the prognosis of pancreatic cancer patients(P<0.05).Conclusion The expression of ADNP is up-regulated in pancreatic cancer,which is significantly correlated with highly aggressive biological behavior and poor prognosis.ADNP can be utilized to assess the occurrence and progression of pancreatic cancer.
