Expression and biological function of Y-box binding protein 1 in gastric cancer

Abstract：Objective To investigate the effect of Y-box binding protein 1(YB-1)on the malignant behavior of gastric cancer cells.Methods The expression of YB-1 in 384 gastric cancer tissues and 35 adjacent normal tissues was analyzed by the cancer genome atlas(TCGA)database,and its level and prognosis in gastric cancer were emphatically analyzed.To select sequences with higher knockdown efficiency against YB-1,siYB-1#2 and siNC were transfected into gastric cancer AGS and SGC-7901 cell lines.After targeting YB-1 gene knockdown and overexpression in gastric cancer cell lines AGS and SGC-7901,the effects of YB-1 on cell proliferation,apoptosis,migration,and invasion were detected by MTT assay,colony formation assay,flow cytometry,and Transwell chamber assays.Results TCGA data revealed that the level of YB-1 in gastric cancer was abnormally elevated compared to adjacent normal tissues(P<0.001),and there was a positive correlation between YB-1 expression and poor prognosis.The results of MTT assay and clone cell formation experiment showed that compared with the siNC group,the cell proliferation ability of siYB-1#2 group was significantly reduced(P<0.05),and the number of cell clones was significantly reduced(P<0.05).The results of flow cytometry detection showed that the apoptosis rate of SGC-7901 gastric cancer cells after knocking down YB-1 was(15.27±0.75)%,significantly higher than that of the control group(8.43±0.51)%,and the difference was statistically significant(P<0.05).The Transwell experiment results showed that compared with the siNC group cells,the number of cells passing through the Transwell chamber in the siYB-1#2 group decreased,indicating that knocking down YB-1 could significantly inhibit the migration and invasion ability of AGS and SGC-7901 cells(P<0.05).Conversely,overexpression of YB-1 promoted cell proliferation,enhanced colony formation(P<0.05),and increased migration and invasion capabilities(P<0.05).Conclusion YB-1 may be involved in the development,progression,and metastasis of gastric cancer.
