Level and clinical significance of calcium activated nucleotidase 1 in colorectal cancer tissues

Abstract:Objective To elucidate the expression pattern of calcium activated nucleotidase 1(CANT1)in colorectal cancer tissues and explore its potential value as a prognostic molecular marker.Methods One hundred and twenty-seven patients with primary colorectal cancer admitted between January 2019 and January 2021 were included consecutively.All cases were confirmed by pathology and paired cancer tissue and adjacent normal tissue specimens were obtained.By using real-time fluorescence quantitative PCR to quantitatively detect the expression level of CANT1 mRNA,patients were divided into a low expression group and a high expression group based on the median value of the detection results.Pearson chi square test was used to analyze the correlation between CANT1 expression level and clinical pathological characteristics of colorectal cancer.Kaplan-Meier method was used to plot survival curves,Log rank test was used to compare survival differences between groups,and a multivariate Cox proportional hazards regression model was constructed to evaluate prognostic factors.Results The expression level data of CANT1 in cancer tissue and adjacent tissues followed a normal distribution as confirmed by D'Agostino Pearson normality test(P>0.05).Quantitative analysis showed that the expression level of CANT1 in cancer tissues(4.445±1.091)was significantly higher than that in adjacent normal tissues(1.350±0.349),and the difference was statistically significant(t=30.445,P<0.001).Stratified analysis revealed that tissue CANT1 levels were not related to gender,age,family history,tumor location,differentiation degree,N stage and carcinoembryonic antigen level(P>0.05),but were related to tumor size,T stage and TNM stage(P<0.05).The median disease-free survival of patients with low level of CANT1 was not achieved,which was better than that of patients with high level at 54 months(P=0.002).Multivariate analysis showed that elevated level of CANT1 was an independent risk factor for recurrence in colorectal cancer patients(HR=2.055,95%CI:1.208~3.496,P=0.008).Conclusion CANT1 is significantly upregulated in colorectal cancer,and its expression level is closely related to tumor progression and poor prognosis,suggesting that CANT1 may serve as a novel molecular marker and potential therapeutic target.
