Expression and clinical significance of BolA family member 2 in colorectal cancer tissues

Abstract：Objective To investigate the expression and clinical significance of BolA family member 2(BOLA2)in colorectal cancer tissues.Methods The expression of BOLA2 in colorectal cancer tissues from the TCGA database was analyzed based on the UALCAN website.Immunohistochemistry and real-time quantitative PCR were used to detect the expression levels of BOLA2 in colorectal cancer tissues and adjacent normal tissues.The relationship between BOLA2 expression and the clinicopathological features as well as the prognosis of colorectal cancer was analyzed.Multivariate analysis was performed using the Cox proportional hazards regression model.Results Online analysis results showed that the BOLA2 level[median(IQR)]in 286 colon cancer tissues was 115.14(97.157-145.736),significantly higher than 80.482(75.660-93.820)in 41 normal tissues.Similarly,the BOLA2 level in 166 rectal cancer tissues was 158.980(104.723-241.041),significantly higher than 76.605(71.729-80.835)in 10 normal tissues,with both differences being statistically significant(P<0.001).In 80 colorectal cancer tissues,the integrated optical density value,positive rate and mRNA level of BOLA2 were 2.262±0.357,42.5%(34/80)and 4.512±1.974,respectively,all significantly higher than 1.001±0.464,26.25%(21/80)and 1.005±0.772 in adjacent normal tissues with statistically significant differences(P<0.001).Kaplan-Meier survival curve revealed that the median overall survival of BOLA2-negative patients was not reached,which was longer than 34.5 months of BOLA2-positive patients(HR=4.588,95%CI:2.019-10.426,P<0.001).Further multivariate analysis confirmed that BOLA2-positive expression was an independent risk factor for poor prognosis in colorectal cancer patients(HR=4.197,95%CI:2.144-8.216,P=0.007).Conclusion High expression of BOLA2 contributes to the malignant progression and poor prognosis of colorectal cancer,suggesting its potential as a therapeutic and diagnostic target for this disease.
