Relationship between FANCD2 and HSBP1 expression levels and pathological features/prognosis in thyroid cancer patients

Abstract:Objective To investigate the relationship between Fanconi anemia complementation group D2 protein(FANCD2)and heat shock factor binding protein 1(HSBP1)expression levels and pathological features/prognosis in thyroid cancer patients.Methods A total of 88 patients with thyroid cancer from January 2019 to March 2022 were selected to test the mRNA and protein expression levels of FANCD2 and HSBP1 via immunohistochemistry and quantitative real-time polymerase chain reaction.Relationships between the FANCD2 and HSBP1 expression levels and the clinical pathological characteristics of thyroid cancer were analyzed.Pearson correlation analysis was applied to determine the correlation between FANCD2 and HSBP1 protein expression.The prognostic significance was assessed using recurrence data from follow-up,with Kaplan-Meier survival curves being constructed to examine the association between FANCD2/HSBP1 expression levels and clinical outcomes in thyroid cancer.Results The expression levels of HSBP1 and HSBP1 in thyroid cancer tissues were higher than those in paracancer tissues(P<0.05).There were 56 cases with high expression of FANCD2 and 32 cases with low expression of FANCD2,54 cases with high expression of HSBP1 and 34 cases with low expression of HSBP1.Individuals with high expression of FANCD2 and HSBP1 have more advanced TNM staging and higher lymph node metastasis rates(P<0.05).Pearson correlation analysis showed that the proportion of FANCD2 and HSBP1 positive expression cells was positively correlated(r=0.853,P<0.001).After 24 months of follow-up observation,a total of 8 out of 88 patients experienced recurrence,with a recurrence rate of 9.09%.The recurrence rate of patients with HSBP1 and FANCD2 high expression was significantly higher than those with low expression at 24 months(P<0.05).Conclusion The high expression of FANCD2 and HSBP1 may promote the malignant progression of thyroid cancer by inhibiting ferroptosis,and their value as prognostic markers and potential therapeutic targets deserves further exploration.
