Identification of serine protease 35 as a novel prognostic biomarker in glioma and its tumor-suppressive functional study

Abstract:Objective To investigate the expression of serine protease 35(PRSS35)in glioma and its prognostic relevance and verify its effect on the proliferative capacity of glioma cells.Methods The key differential genes of glioma were analyzed from Chinese Glioma Genome Atlas and The Cancer Genome Atlas databases by machine learning methods,and their prognostic relevance to glioma patients was analyzed by machine learning algorithms.Human astrocytes and glioma cell lines were cultured in vitro,and the expression levels of PRSS35 were detected using quantitative polymerase chain reaction and Western blot.The PRSS35 expression levels were altered using transfection.Cell counting kit-8 and terminal deoxynucleotidyl transferase dUTP nick end labeling were used to detect glioma cell viability and apoptosis levels.Western blot was used to detect the expression levels of apoptosis-related proteins in glioma cells.Results PRSS35 expression was decreased in glioma with high malignancy,and the prognosis level of glioma patients with low expression of PRSS35 was poor.Overexpression of PRSS35 was able to inhibit the proliferation of glioma cells and promote their apoptosis level.Conclusion The low expression of PRSS35 in glioma suggests poor prognosis for patients.Overexpression of PRSS35 may inhibit glioma cell proliferation by regulating apoptosis-related protein expression,indicating that PRSS35 could be a potential therapeutic target for glioma.
