Effects of long intergenic non-protein coding RNA 1089 on migration and invasion of cervical cancer cells

Abstract：Objective To characterize the mechanisms and functions of long intergenic non-protein coding RNA 1089(LINC01089)on migration and invasion of cervical cancer cells.Methods Quantitative real-time PCR(qPCR)was used to evaluate the expressions of LINC01089 and microRNA-346(miR-346)in normal cervical epithelial cells End1/E6E7 and cervical cancer cells(CaSki,SiHa,C-33A and HeLa).HeLa cells were cultured and divided into pcDNA3.1 group,pcDNA3.1-LINC01089 group(over-expression of LINC01089)and pcDNA3.1-LINC01089+miR-346 mimics group(over-expression of LINC01089 and miR-346).CCK-8 assay was used to detect cell proliferation.Cell scratch and Transwell experiments were used to evaluate cell migration and invasion.Dual luciferase reporter gene experiment validated the targeting between miR-346 and LINC01089 or RNA binding motif protein X-linked(RBMX).Expression of RBMX,matrix metalloproteinase(MMP)-2 and MMP-9 were detected by qPCR and Western blot.Results LINC01089 expression was lower but miR-346 expression was higher in cervical cancer cells,compared to End1/E6E7 cells(P<0.05).Function analysis verified that ectopic overexpression of LINC01089 inhibited cell abilities of proliferation,migration and invasion,and weakened the expressions of MMP-2 and MMP-9 in HeLa cells(P<0.05).Mechanistically,LINC01089 could bind directly to miR-346,and RBMX was found to be a downstream target gene of miR-346.Moreover,miR-346 mimics partly reverse the suppressive effects of LINC01089 on HeLa cells(P<0.01).Conclusion LINC01089 suppressed proliferation,migration and invasion of cervical cancer cells through regulating the miR-346/RBMX axis,indicating that LINC01089 may be a potential therapeutic target in cervical cancer.
