Advances in molecular mechanisms of immunogenic cell death

Abstract:Immunogenic cell death(ICD),as a specific form of regulated cell death,holds a crucial position in tumor immunotherapy.This review emphasizes the classical pathways of ICD,encompassing immunogenic apoptosis and necrosis.The current known clinical application of anticancer ICD inducers and the experimental stage of anticancer ICD inducers were analyzed in depth,and the characteristics of ideal ICD inducers were summarized from multiple dimensions.Simultaneously,the molecular signaling pathways through which ICD activates the maturation of dendritic cells and triggers the T lymphocyte-mediated antitumor immunity in the tumor microenvironment were profoundly explored,clarifying the key role of reactive oxygen species(ROS)therein.The intrinsic mechanisms of tumors exert a significant influence on the tumor microenvironment,tumor development,and treatment responses,and ICD brings hope for restoring the immunogenicity of"cold"tumors.Intensive research on ICD is beneficial for promoting the development of novel strategies for tumor treatment and possesses significant theoretical and practical implications.
