Expression of SNX10 in cervical cancer and its association with prognosis and immune infiltration

Abstract:Objective To investigate the expression of sorting nexin 10(SNX10)in cervical cancer and its relationship with prognosis and immune infiltration.Methods Data on cervical cancer and adjacent tissues were obtained from The Cancer Genome Atlas(TCGA)database and the high-throughput gene expression(GEO)database.The expression level of SNX10 in cervical cancer was analyzed using R software.The potential of SNX10 as a novel biomarker for the auxiliary diagnosis of cervical cancer was analyzed by ROC curve.The relationship between SNX10 expression and prognosis was analyzed by Kaplan-Meier method.Gene enrichment analysis was conducted to explore the potential role of SNX10 in cervical cancer.Correlation tests were performed to analyze the correlation between SNX10 expression and tumor microenvironment,tumor immune cell infiltration and immune checkpoints.Additionally,52 cases of cervical cancer tissues were collected from January 2018 to December 2020,and the expression level of SNX10 was detected by immunohistochemistry.Results Database analysis showed that the expression level of SNX10 in cervical cancer tissues was higher than that in adjacent tissues(P<0.05).Survival analysis showed that the patients with high expression of SNX10 had shorter overall survival.Immunohistochemical staining showed that the positive expression rates of SNX10 in cervical cancer tissues and adjacent tissues were 69.2%and 38.5%respectively(P=0.0017).The area under the curve of SNX10 for diagnosing cervical cancer in GSE63514,GSE9750 and GSE7803 datasets were 0.893,0.877 and 0.884 respectively.GO analysis and GSEA pathway enrichment analysis showed that SNX10 might be involved in the activity of the immune microenvironment in cervical cancer.The TISCH online website tool showed that SNX10 was enriched in macrophages,and the higher the expression of SNX10 in cervical cancer,the higher the expression of CD163 and the more M2 macrophage infiltration.SNX10 was positively correlated with the expression of immune checkpoints PDCD1(r=0.461),CD274(r=0.422),TIGIT(r=0.427),LAG3(r=0.455)and HAVCR2(r=0.572)with significance(P<0.05).Conclusion SNX10 may be a potential diagnostic and prognostic marker for cervical cancer,providing new clues for improving the diagnosis and treatment of cervical cancer.
