Exploration of the key genes and clinical significance of oxaliplatin-induced peripheral neurotoxicity based on bioinformatics

Abstract:Objective Toexplore the hub gene expression and clinical significance of oxaliplatin-induced peripheral neurotoxicity(OIPN)by bioinformatics analysis.Methods Bioinformatics analysis was used to identify the differential expression genes,and the enrichment of differential expression genes were investigated.Least Absolute Shrinkage and Selection Operator and Support Vector Machine-Recursive Feature Eliminationalgorithms were used to identify the hub genes from the differential expression genes,and the receiver operating characteristic curve analysis was used to evaluate their diagnostic efficacy.Survival prognosis was analyzed using colon cancer patients from public databases.Results Eighty-one differential expression genes that induced peripheral neurotoxicity after oxaliplatin treatment in colon cancer.They were associated with the innate immune response.Moreover,radical S-adenosyl methionine domain containing 2(RSAD2)and steroid 5 alpha-reductase 2(SRD5A2)were identified as hub genes inducing OIPN.SRD5A2 was inversely associated with the survival prognosis of colon cancer patients.Conclusion RSAD2 and SRD5A2 are deemed as the hub genes for peripheral neurotoxicity after oxaliplatin treatment,and their regulatory networks provide new ideas for the pathogenic mechanism and diagnosis of OIPN.
