Effects of luteolin regulating autophagy on proliferation,apoptosis,invasion and migration of colon cancer cells

Abstract:Objective To explore the effects of luteolin regulating autophagy on proliferation,apoptosis,invasion and migration of colon cancer cells.Methods HT-29 and SW620 cells were assigned into normal group,luteolin group,and luteolin and mammalian target of rapamycin(mTOR)agonist group.Normal group was cultured routinely,while cells in the luteolin group were treated with 20 μmol/L luteolin intervention under normal culture conditions;Cells in the luteolin and mTOR agonist group were exposure to a combination of 20 pmol/L luteolin and 5 μmol/L mTOR agonist.MTT,flow cytometry and Transwell assays were used to detect cell abilities of proliferation,apoptosis,and invasion and migration.Cellular ultrastructure was observed by using transmission electron microscopy.Western blot was used to detect expressions of autophagy-related proteins including microtubule associated protein light chain 3(LC3),Beclin1(BECN1)and autophagy related gene 7(ATG7).Results Compared with the normal group,there were decreased proliferation rate,increased apoptotic rate and autophagosome,and the reduced number of invasive and migratory cells together with increased LC3Ⅱ/LC3I ratio and expressions of BECN1 and ATG7 in HT-29 and SW620 cells in the luteolin group(P<0.05).However,after simultaneous intervention with mTOR agonist,compared with the luteolin group,the luteolin and mTOR agonist group showed restoration of proliferation,invasion,and migration abilities,while autophagy levels decreased.Conclusion Luteolin inhibits the biological activity of colon cancer cells possibly by activating autophagy.
