Expression of long non-coding RNA DLEU7 antisense RNA 1 in gastric cancer and its effect on cell migration and invasion

Abstract：Objective To illustrate the biological role of DLEU7 antisense RNA 1(DLEU7-AS1)in the tumorigenesis of gastric cancer.Methods Online analysis of the expression of DLEU7-AS1 in gastric cancer tissues was performed.DLEU7-AS1 expression in gastric cancer MGC-803 cells were measured by quantitative real-time PCR(qPCR).Small interfering RNA(siRNA)sequences targeting DLEU7-AS1(siRNA-DLEU7-AS1)and negative control sequence(siRNA-NC)were constructed and transfected into MGC-803 cells.Then,cells were divided into blank control group(no treatment),negative control(NC)group(cells transfected with siRNA-NC),and si-DLEU7-AS1 group(cells transfected with siRNA-DLEU7-AS1 sequence).MTT,scratch-healing and Transwell assays were used to assess cell proliferation,migration and invasion.Then,qPCR and Western blot were used to detect the expression levels of epithelial-mesenchymal transition related factors including N-cadherin(N-cad),E-cadherin(E-cad)and fibronectin(FN).Results The DLEU7-AS1 expression was upregulated in gastric cancer tissues,and its expression was higher in MGC-803 cells than in GES-1 cells(P＜0.01).Compared with the NC group,the proliferation activity,scratch healing rate,and number of membrane penetrations of MGC-803 cells in the si-DLEU7-ASl group decreased,while the expression levels of N-cad and FN decreased,and the expression level of E-cad increased,with statistical significance(P＜0.01).There was no statistically significant difference in the above indicators between the blank control group and the NC group(P＞0.05).Conclusion DLEU7-AS1 may play an oncogenic role in proliferation,migration,invasion and epithelial-mesenchymal transition of gastric cancer cells.This could be a potential therapeutic target for gastric cancer.
