Effects of solute carrier family 10 member 4 on the proliferation and mitochondrial autophagy of osteosarcoma U2OS cells

Abstract:Objective To investigate the expression characteristics of solute carrier family 10 member 4(SLC10A4)in osteosarcoma and its effect on the proliferation and mitochondrial autophagy of U2OS cells.Methods The Cancer Genome Atlas database was used to analyze the expression level of SLC10A4 in osteosarcoma tissues and its relationship with survival prognosis.Changes in cell proliferation ability and mitochondrial autophagy related indicators were observed after silencing of SLC10A4 in U2OS cells.SLC10A4 silence and overexpression of BCL2 interacting protein 3(BNIP3)were performed simultaneously to observe whether it could reverse the effect of SLC10A4 silence on cell proliferation.Results The results of database showed that the expression of SLC10A4 in osteosarcoma tissues was significantly increased,and high expression was associated with poor prognosis(P<0.001).SLC10A4 silencing inhibited cell viability and mitochondrial membrane potential,increased mitochondrial quantity,reduced autophagosome formation,and decreased the expression of mitochondrial autophagy related proteins in U2OS cells.Overexpression of BNIP3 partially reverses the inhibitory effect of SLC10A4 on U2OS cell proliferation.Conclusion High expression of SLC10A4 in osteosarcoma indicates a poor prognosis;silencing SLC10A4 inhibits U2OS cell proliferation by BNIP3-mediated mitochondrial autophagy.
