Progress of ALK-TKIs in neoadjuvant and adjuvant targeted therapy for non-small cell lung cancer

Abstract:Patients with non-small cell lung cancer(NSCLC)have achieved satisfactory efficacy with oncogene-driven targeted therapy and non-oncogene-driven immune checkpoint inhibitors.Among these treatments,tyrosine kinase inhibitors(TKIs)have significantly prolonged survival outcomes and improved quality of life for patients with advanced NSCLC who have the anaplastic lymphoma kinase(ALK)gene mutation.It is currently difficult to harmonize neoadjuvant and postoperative adjuvant treatment regimens due to the paucity of relevant clinical studies and the low prevalence of ALK positivity in NSCLC patients.As a result,it is not possible to draw definitive conclusions about whether patients with early ALK-positive NSCLC can continue to benefit from ALK-TKIs.This article reviews the molecular structure and clinical baseline characteristics of ALK,as well as the major research advances in early ALK-positive NSCLC receiving targeted agents in neoadjuvant and adjuvant therapy.
