Expression and clinical significance of TINCR in prostate cancer tissues

Abstract：Objective To discuss the expression and clinical significance of tissue differentiation-inducing non-protein coding RNA(TINCR)in prostate cancer tissues.Methods Seventy-two pairs of prostate cancer tissues and adjacent tissues surgically removed from our hospital from May 2017 to July 2019 were collected.In situ hybridization(ISH)was used to detect the positive expression of TINCR and an integrated optical density(IOD)was calculated.Real-time quantitative PCR was used to detect the relative expression of TINCR and then we analyzed its relationship with the clinical pathological characteristics and prognosis of prostate cancer.Kaplan-Meier method was used for survival analysis,while the Cox proportional risk model was used for multivariate analysis.Results Compared with adjacent tissues,the IOD value and expression level of TINCR in prostate cancer tissues were reduced(P<0.05).The TINCR expression was not related to the age,PSA level and lymph node metastasis of prostate cancer patients(P>0.05),but was related to clinical T stage,Gleason score and TNM stage(P<0.05).TINCR was lowly expressed in tissues with clinical T3+ T4 stage,Gleason score greater than 8,and TNM Ⅲ+Ⅳ stage.Univariate analysis showed that the TINCR expression was associated with overall survival(OS)in patients with prostate cancer.Among them,the median OS in patients with high TINCR expression was not reached,better than 48.0 months in patients with low TINCR expression(Log-rank χ2=3.954,P=0.047).Multivariate analysis showed that decreased expression of TINCR was a risk factor for poor prognosis of prostate cancer(HR= 4.953,95%CI:1.576-15.565,P=0.006).Conclusion Downregulation of TINCR expression in prostate cancer tissues can serve as a potential molecular marker for the diagnosis and prognosis prediction of prostate cancer patients.
