Correlation analysis of circRNAs regulatory network in non small-cell lung cancer tissues

Abstract:Object4ive To explore the role of differentially expressed circular RNAs(circRNAs)in non small-cell lung cancer(NSCLC)tissues as to find significant tumor markers.Methods Expression data of circRNAs and miRNAs in NSCLC tissues were obtained by using GEO database and analyzed with R software.Then,miRanda and RNAhybrid software were employed to predict the binding miRNAs of circRNAs.After intersection with differentially expressed miRNAs,a circRNAs-miRNAs interaction network diagram was constructed.Expressions of their parent genes in lung squamous cell carcinoma tissues were analyzed using the TCGA database,and survival analysis was perform using Kaplan-Meier Plotter software.Results Through the combined analysis of the two GEO databases including GSE101586 and GSE112214,a total of 148 differentially expressed circRNAs were found,of which 14 up-regulated circRNAs and 134 down-regulated circRNAs.Through GEO analysis,there were 975 differentially expressed miRNAs in NSCLC tissues,and 70 miRNAs coincided with the miRNAs predicted by the top 8 fold change circRNAs.From TCGA tissue expression analysis,it was found that the expressions of PDK1 and CYP24A1 increased in lung squamous cell carcinoma,and the expressions of LIFR and NTNG1 decreased,consistent with the expression trend of circRNAs.Survival analysis found that high expression of GCLC,PDK1 and NTNG1 was closely associated with poor overall survival in patients with lung squamous cell carcinoma,whereas LIFR and KLHL20 were the opposite.Conclusion Differential expression circRNAs in NSCLC tissues will be meaningful tumor markers.
