Effects of β-elemene regulating HULC expression on malignant biological behavior of hepatocellular carcinoma cells

Abstract:Objective To investigate the role of hepatocellular carcinoma up-regulated long non-coding RNA(HULC)in the regulation of malignant biological behavior of hepatocellular carcinoma cells by β-elemene.Methods Bioinformatics analysis of HULC expression in hepatocellular cancer tissues was performed using TCGA database.HULC expression in hepatocellular cancer cells was measured by real-time quantitative PCR.SMMC-7721 cells were transfected with HULC overexpression plasmid pcDNA3.1-HULC or small interfering RNA si-HULC.CCK-8 method was used to evaluate the effects of β-elemene and HULC on the cell proliferation.Changes in cell migration and invasion abilities were analyzed via scratch and Transwell experiments.Western blot was used to detect the expression of factors related to epithelial-mesenchymal transition(EMT).Results HULC expression was upregulated in hepatocellular carcinoma tissues and cells.Knocking-down HULC or β-elemene treatment could inhibit the EMT process and abilities of proliferation,migration and invasion of SMMC-7721 cells.In addition,β-elemene treatment reduced HULC expression,while HULC overexpression partly blocked the inhibitory effects of β-elemene on EMT process and abilities of proliferation,migration and invasion.Conclusion β-Elemene can regulate the malignant biological behavior of hepatocellular carcinoma cells by downregulating HULC expression.It provides new insights into its potential application of targeting HULC by β-elemene as a treatment for hepatocellular carcinoma.
