Effects of LIPF on the proliferation,migration and invasion of gastric cancer cells by regulating MAPK signaling pathway

Abstract:Objective To clarify the role of lipase F,gastric type(LIPF)on the proliferation,migration and invasion of gastric cancer cells and its impact on mitogen activated protein kinase(MAPK)signaling pathway.Methods GEPIA and Human Protein Altas were used to investigate the LIPF expression in gastric cancer tissues.Moreover,qPCR and Western blot were applied to check the expression of LIPF in gastric cancer cells SGC-7901.SGC-7901 cells were divided into Negative control(NC)group,LIPF overexpression group and LIPF overexpression+MAPK activator group.Cell viability,migration and invasion in SGC-7901 cells were analyzed by MTT,scratch and Transwell assays.Additionally,qPCR and Western blot were used to examine expressions of cyclin dependent kinase 4(CDK4),matrix metallopeptidase 9(MMP9),phosphorylated p38(p-p38)and phosphorylated c-Jun amino terminal kinase(p-JNK).Results LIPF expressions were significantly depleted in gastric cancer tissues and SGC-7901 cells(P<0.05).Compared with NC group,the cell viability,healing rate and number of invaded cells were attenuated in LIPF overexpression group(P<0.05).Downregulation of CDK4,MMP9,p-p38 and p-JNK were observed in LIPF overexpression group compared to NC group(P<0.05).Additionally,compared with LIPF overexpression group,the cell viability,healing rate and number of invaded cells were increased in LIPF overexpression+MAPK activator group(P<0.05).Expressions of CDK4,MMP9,p-p38 and p-JNK were promoted in LIPF overexpression+MAPK activator group than those in LIPF overexpression group(P<0.05).Conclusion LIPF could suppress proliferation,migration and invasion of gastric cancer through regulating p38 MAPK signaling pathway in vitro,which may offer a new therapeutic intervention for gastric cancer patients.
