Effects of long non-coding RNA EMX2OS on proliferation,migration and invasion of cervical cancer cells through miR-150/TP53 axis

Abstract：Objective To explore the role of EMX2 opposite strand/antisense RNA(EMX2OS)in proliferation,migration and invasion of cervical cancer cells through targeting microRNA-150(miR-150)/tumor protein p53(TP53)axis.Methods Real-time quantitative polymerase chain reaction(qPCR)was used to detect levels of EMX2OS and miR-150 in cervical cancer cells.C-33A cells were transfected with EMX2OS overexpression vector pcDNA3.1-EMX2OS or miR-150 mimics,and divided into 4 groups:pcDNA3.1 group,pcDNA3.1-EMX2OS group(overexpression of EMX2OS),pcDNA3.1-EMX2OS+miR-NC group and pcDNA3.1-EMX2OS+miR-150 mimics group(overexpression of EMX2OS and miR-150).Cell growth,migration and invasion were monitored by MTT,wound-healing and Transwell assays.The interaction relationships between EMX2OS/miR-150 and miR-150/TP53 were analyzed by dual-luciferase reporter.Results EMX2OS was decreased in cervical cancer cells,while miR-150 was increased(P<0.05).Compared with pcDNA3.1 group,the cell proliferation activity,healing rate,the number of invaded cells and miR-150 expression were curtailed in pcDNA3.1-EMX2OS group,while the expression of TP53 was promoted(P<0.05).Compared with pcDNA3.1-EMX2OS+miR-NC group,the cell proliferation activity,healing rate,the number of invaded cells and miR-150 expression were elevated in pcDNA3.1-EMX2OS+miR-150 mimics group,while the expression of TP53 was declined(P<0.05).Both EMX2OS and TP53 bounded to miR-150.Conclusion EMX2OS suppressed cell growth,migration,and invasion possibly via the miR-150/TP53 axis in cervical cancer,which could be a potential target for treating cervical cancer patients.
