The expression and clinical significance of TRPV1 and TRPA1 in gastric cancer

Abstract：Objective To investigate the expression and clinical significance of TRPV1 and TRPA1 in gastric cancer tissue.Methods Ninety gastric cancer tissue samples and corresponding adjacent tissue samples were collected from patients who underwent surgical resection in our hospital from January 2013 to April 2017.Apply real-time fluorescence quantitative polymerase chain reaction to detect the expression of TRPV1 and TRPA1 in the above samples,and analyze their relationship with clinical pathological characteristics and prognosis of patients.The efficacy of both method in diagnosing gastric cancer was analyzed using the receiver operating curve(ROC).Kaplan-meier method was used to draw survival curves.The Cox risk proportional regression model was used to analyze the factors affecting the prognosis of gastric cancer.Results The expression level of TRPV1 in gastric cancer tissue was 3.353±1.007,which was higher than 1.484±0.557 in adjacent tissues,with a statistically significant difference(P＜0.05).The expression level of TRPA1 in gastric cancer tissue was 3.135±1.058,which was higher than 1.466±0.600 in adjacent tissues,with a statistically significant difference(P＜0.05).Spearman rank correlation analysis showed a positive correlation between TRPV1 and TRPA1 levels in gastric cancer tissue(r=0.423,P＜0.05).ROC curve analysis showed that the area under the curve for diagnosing gastric cancer at TRPV1 level was 0.933,with sensitivity and specificity of 82.2%and 95.6%,respectively.The area under the curve for diagnosing gastric cancer at the TRPA1 level was 0.896,with sensitivity and specificity of 85.6%and 87.8%,respectively.The expression levels of TRPV1 and TRPA1 are related to TNM staging,differentiation degree,and infiltration depth.The median OS of patients with high expression of TRPV1 was 25.0 months,less than 58.0 months of patients with low expression(P＜0.05).The median OS of patients with low expression of TRPA1 was 29.0 months,less than 56.0 months in patients with low expression(P＜0.05).The Cox risk proportional regression model shows that TRPV1 expression,TRPA1 expression,TNM staging,differentiation degree,and infiltration depth are independent factors affecting the prognosis of gastric cancer patients.Conclusion The expression of TRPV1 and TRPA1 is significantly increased in gastric cancer tissue,and the high expression of TRPV1 and TRPA1 is related to the prognosis of patients,which can serve as an independent factor for judging the prognosis of gastric cancer patients.
