Expression and clinical significance of long chain non coding RNA DLEU7-AS1 in renal cell carcinoma

Abstract:Objective To investigate the expression and clinical significance of long chain non coding RNA DLEU7-AS1 in renal cell carcinoma tissue.Methods One hundred and twelve renal cell carcinoma tissue samples and 94 corresponding normal tissue samples were selected,and serum samples were collected from 112 patients and 50 normal healthy volunteers.The real-time fluorescence quantitative PCR(qPCR)method was used to detect the expression of DLEU7-AS1,analyze the relationship between DLEU7-AS1 expression and clinical pathological characteristics of renal cell carcinoma,and analyze the diagnostic efficacy of DLEU7-AS1 using subject operating characteristic(ROC)curves.Analyze the relationship between DLEU7-AS1 and the prognosis of renal cell carcinoma through the TCGA database.Kaplan-meier method was used to draw survival curves.Results The result were analyzed using 523 renal tumor tissue samples and 72 normal tissue samples from the TCGA database,and DLEU7-AS1 was significantly upregulated in tumors(P<0.05).The qPCR result showed that the expression level of DLUE7-AS1 in renal cancer tissue was 4.16±1.91,which was higher than 1.56±0.81 in adjacent tissues,with a statistically significant difference(P<0.05).The expression of DLEU7-AS1 was correlated with histological grading,TNM staging,T staging,and M staging(P<0.05).The area under the curve(AUC)for ROC diagnosis of renal cell carcinoma was 0.856(95%CI:0.783-0.929;P<0.01),with sensitivity and specificity of 87.65%and 82%,respectively.The TCGA database shows that patients in the low expression group of DLEU7-AS1 and the high expression group have significantly longer disease-free survival(P=0.039)and overall survival(P=0.041).The medical records of our hospital show that compared with patients in the high expression group,patients in the low expression group have a longer progression free survival(P<0.05)and overall survival(P<0.05).Conclusion DLEU7-AS1 is highly expressed in renal cell carcinoma and can serve as a novel diagnostic and prognostic biomarker for renal cell carcinoma.
