Experimental study of vitexin on the regulation of proliferation and migration of lung adenocarcinoma A549 cells by the NF-κB/MMP-9 pathway

Abstract:Objective To investigate the effects and possible mechanisms of vitexin on the proliferation and migration of lung adenocarcinoma A549 cells.Methods A549 cells were treated with different concentrations of vitexin(0,10,20,40,60,80,100 μmol/L),and 0.1%dimethyl sulfoxide(DMSO)was used as the solvent control group.The cell survival rate of each group was measured using the CCK-8 method,and the half inhibitory concentration(IC50)was calculated.According to IC50,vitexin was divided into low,medium,and high concentration groups.The cell migration ability of each group was detected by scratch test,and the expression level of nuclear factor B(NF-κB)signaling pathway related proteins in each group was detected by Western blotting method.The effect of vitexin on the transcriptional activity of matrix metalloproteinase-9(MMP-9)in A549 cells was determined by luciferase assay.Results Compared with the blank control group,after 24 hours of treatment with different concentrations of vitexin(10,20,40,60,80,100 μmol/L),the cell viability of A549 decreased with the increase of vitexin concentration(P<0.05)in a concentration dependent manner,with an IC50 of(38.93±2.15)μmol/L;The 24-hour cell migration rates of the low concentration group(20 μmol/L),medium concentration group(40 μmol/L),and high concentration group(60 μmol/L)were(19.30±1.25)%,(15.48±1.30)%,and(12.03±0.95)%,respectively,lower than the blank control group's(25.05±1.50)%,with statistical significance(P<0.05).Compared with the blank control group,the expression of IKK β,p-IKK β,NF-κB p65,and MMP-9 proteins in different concentration groups of vitexin was significantly reduced(P<0.05)in a concentration dependent manner.The result of luciferase assay showed that different concentrations of vitexin significantly inhibited the promoter activity of MMP-9.Conclusion Vitexin can inhibit the proliferation and migration of A549 cells,and its mechanism may be related to downregulating the expression of NF-κB signaling pathway related proteins,leading to a decrease in downstream MMP-9 protein expression.
