Serum level and clinical significance of monoglyceride lipase in patients with oral squamous cell carcinoma

Abstract:Objective To investigate the serum level and clinical significance of monoglyceride lipase ( MGLL) in patients with oral squamous cell carcinoma (OSCC). Methods Gene Expression Profiling Interactive Analysis (GEPIA) was used to analyze the expression patterns of MGLL in 33 kinds of cancer tissues. From January 2013 to December 2016, serum samples of 94 patients with OSCC and 96 physical examinees were collected. The quantitative real-time PCR (QPCR) was used to detect the MGLL level in the above serum samples and receiver operating characteristic (ROC) curve was applied to evaluate the efficacy of serum MGLL level in diagnosis of OSCC. According to the mean value of serum MGLL level, the patients were divided into low level (≤mean) and high level (> mean). The relationship between serum MGLL levels and clinicopathological characteristics and prognosis was analyzed. Cox multivariate risk model was used to analyze the prognostic factors of OSCC. Results GEPIA on-line analysis showed that the expres-sion of MGLL was abnormal in 15 kinds of tumors, among which 5 kinds were increased and other 10 kinds were decreased. The medi-an level of MGLL in squamous cell carcinoma of head and neck was 19. 4, lower than 65. 94 in normal tissues (P<0. 05). The results of QPCR showed that the serum MGLL levels of patients with OSCC was 0. 471±0. 264, lower than 1. 501±0. 687 of the physical exam-inees (P<0. 05). The area under curve of ROC showed that the serum MGLL level was 0. 933 (95% CI: 0. 897-0. 968). When selec-ting 0. 903 as the diagnostic threshold, the yoden index was the highest (0. 7595) with the sensitivity of 95. 74% and the specificity of 80. 21%. MGLL level was not related to age and gender, but related to tumor size, smoking history, pathological grade, lymph node metastasis and TNM stage (P<0. 05). The median overall survival of patients with low MGLL level was 22. 5 months, lower than that of patients with high level (P<0. 05). Multivariate analysis showed that MGLL level was an independent factor affecting the prognosis of OSCC (OR=1. 355, 95%CI: 1. 115-1. 647). Conclusion The serum level of MGLL in OSCC patients is low, which is involved in the occurrence and development of OSCC and is related to poor prognosis. It can be used as a potential index for diagnosis and prognosis evaluation of OSCC.
