Effect of microRNA-1180 on invasion, migration and TGF-β signaling pathway of hepatoma cells

Abstract:Objective To investigate the influence of microRNA-1180 (miR-1180) on invasion, migration and transforming growth factor-β (TGF-β) signaling pathway of hepatoma cells. Methods Real-time quantitative PCR (QPCR) was used to detect the miR-1180 levels of LO2 and hepatoma cells (MHCC97-H, Huh7, SMMC-7721). SMMC-7721 cells were cultured routinely and trans-fected with miR-1180 inhibitor (Inhibitor group) or negative control (NC group) by liposome, and the Control group was treated with-out any transfection. The miR-1180 level, scratch healing rate and number of membrane penetrating cells were measured by MTT color-imetry, scratch test and Transwell test. The relationship between miR-1180 and secreted frizzled related protein 1 ( SFRP1) was con-firmed by bioinformatics and double luciferase reporter gene system. The levels of TGF-β1, matrix metalloproteinase ( MMP)-3 and MMP-9 were detected by QPCR and Western blotting. Results The miR-1180 levels of hepatoma cells were higher than that of LO2 cells (P<0. 05). Compared with Control group and NC group, the miR-1180 level and proliferative activity at 48 and 72 h-transfection were decreased in Inhibitor group (P<0. 05). The wound healing rate and number of transmembrane cells were ( 19. 264±4. 619)%and 133. 863±16. 216 in Inhibitor group, lower than (67. 579±7. 734)% and 316. 906±25. 753 in NC group and (65. 237±5. 621)%and 326. 815±28. 946 in Control group (P<0. 05). Levels of TGF-β1, MMP-3 and MMP-9 in Inhibitor group were lower than those in Control group and NC group, while SFRP1 level in Inhibitor group was higher than that in Control group and NC group (P<0. 05). There was no significant difference between Control group and NC group on the above indicators (P>0. 05). MiR-1180 mimics can re-duce the relative fluorescence intensity of cells transfected with vector carrying wild-type SFRP1 3’ untranslated region (P<0. 05). Conclusion MiR-1180 is highly expressed in hepatoma cells, which may activate TGF-β signal pathway by targeting SFRP1. This miRNA plays a role in promoting the migration and invasion of hepatoma cells, and may be a new and promising therapeutic target for hepatoma.
