Expression and clinical significance of SOX21?AS1 in non?small cell lung cancer tissues

Abstract:Objective To investigate the expression and clinical significance of SOX21 antisense RNA 1 ( SOX21-AS1) in non-small cell lung cancer (NSCLC) tissues.Methods Ninety-four pairs of NSCLC patients??cancer tissues and matched adjacent tissues were collected from September 2014 to June 2017. The levels of SOX21-AS1 in the above tissues were detected by real-time quantitative polymerase chain reaction ( QPCR). The relationship between SOX21-AS1 level and clinicopathological characteristics ( sex, age, smoking history, tumor size, lymph node metastasis, TNM stage and disease) were analyzed.According to the follow-up data, the progno-sis of NSCLC patients with different SOX21-AS1 levels was analyzed. Results The level of SOX21-AS1 in NSCLC tissues was 3. 870± 1. 200, higher than 1. 215±0. 565 in adjacent tissues (P<0. 05).Receiver operating characteristic curve analysis showed that the area un-der curve of tissue SOX21-AS1 level in diagnosis of NSCLC was 0. 986 (95% CI: 0. 975-0. 998), and the Yoden index was the highest (0. 904) when the threshold value was 2. 017 with the sensitivity and specificity of 95. 74% and 94. 68%. The median of SOX21-AS1 in 94 NSCLC tissues was 3. 843, which was divided into 47 cases with low expression (<3. 843) and 47 cases with high expression (>3. 843). The results showed that SOX21-AS1 expression was not related to sex, age and smoking history of NSCLC, but to tumor size, lymph node metastasis, pathological type and TNM stage (P<0. 05).SOX21-AS1 levels were higher in patients with positive nodal metas-tasis, larger tumors and later clinical stages. The median overall survival of high expression of SOX21-AS1 was 31. 5 months, lower than 47. 0 months of low expression (P<0. 05). Multivariate analysis showed that the overexpression of SOX21-AS1 was an independent factor for the poor prognosis of NSCLC ( HR=2. 153, 95% CI: 1. 657-4. 156, P<0. 05). Conclusion SOX21-AS1 is highly expressed in NSCLC tissues and participates in the occurrence and development of NSCLC.The prognosis of NSCLC patients with high level of lncRNA is poor. SOX21-AS1 can be used as a potential molecular diagnostic and prognostic marker for NSCLC.
