Abstract:Traditional treatment such as radiotherapy and chemotherapy for advanced non?small cell lung cancer ( NSCLC) had limited efficacy. In recent years, immune checkpoint inhibitors as a way of active immunotherapy, have opened a new era for the treatment of advanced NSCLC because of their remarkable efficacy and long?lasting response. Programmed cell death?1 ( PD?1) is an immune checkpoint in the immune system that can transmit inhibitory signals when combined with programmed cell death?ligand 1 ( PD?L1) and reduce the effects of cellular immunity. Cancer cells can cause abnormal levels of PD?L1 expression to escape the attack of the immune system. Atezolizumab as an anti?PD?L1 antibody, can bind PD?L1 in advanced NSCLC to prevent it from binding to PD?1, and meanwhile, it activates lymphocyte T cell activity to kill tumor cells in order to achieve the goal of immunotherapy. This article reviews the mechanism and the current clinical application of atezolizumab in clinical trials for advanced NSCLC.
