Expression and clinical significance of microRNA-218-5p in breast cancer tissues

Abstract:Objective To investigate the expression and clinical significance of microRNA-218-5p (miR-218-5p) in breast cancer tissues.Methods From January 2013 to December 2015, 92 pairs of cancerous and paracancerous tissues surgically removed from patients with breast cancer were collected.Real-time quantitative PCR (QPCR) was used to detect the expression of miR-218-5p in the above tissues.The expression difference of miR-218-5p between cancer tissues and adjacent tissues was compared.The relationship between the miR-218-5p expression and clinicopathological characteristics of breast cancer (age, tumor size, lymph node metastasis, TNM stage, menopause, histological grade, ER, PR, HER-2 and Ki-67 expression) were compared.The outpatient or telephone follow-up was used to compare the prognosis of the high expression group (≥mean value) and the low expression group (<mean value) of miR-218-5p level.Results The results of QPCR showed that the expression of miR-218-5p in breast cancer tissues was 0.315±0.150, lower than 1.010±0.298 in adjacent tissues, and the difference was statistically significant (P<0.05). The levels of miR-218-5p in patients with breast cancer were independent of lymph node metastasis, age, menopause, ER expression and PR expression, and significantly related with tumor size, TNM stage, histological grade, HER-2 expression and Ki-67 expression (P<0.05). The patients were followed up for 14 to 60 months with a median follow-up of 45.0 months and a median overall survival (OS) of 57.0months.The median OS of the group with high expression of miR-218-5p was more than 60.0 months, higher than 51.0 months of the group with low expression of miR-218-5p (P<0.05). Univariate analysis also found that the prognosis of breast cancer patients was significantly different from age, tumor size, lymph node metastasis, TNM stage, histological grade and HER-2 expression (P<0.05). Conclusion MiR-218-5p expression is low in breast cancer tissues and has a certain relationship with the occurrence, development and prognosis of breast cancer.The prognosis of patients with low expression of miR-218-5p is poor.This factor has certain potential value in breast cancer screening and prognosis prediction.
