Effects of microRNA-9-3p on invasion and migration of papillary thyroid carcinoma cells by regulating Wnt/β-catenin signaling pathway

Abstract:Objective To investigate the regulation of microRNA-9-3p (miR-9-3p) on Wnt/β-catenin (β-cat) signaling pathway and its effect on the invasion and migration of thyroid papillary carcinoma (PTC) cell line TPC-1.MethodsTPC-1 cells were cultured in vitro and transfected with miR-9-3p inhibitor (inhibitory group) and empty plasmid (empty transfection group), meanwhile blank control group was set up.Real time fluorescence quantitative PCR (QPCR) was used to detect the expression of miR-9-3p in the3 groups.MTT method, scratch test and Transwell cell invasion test were used to detect the proliferation, migration and invasion of the three groups.The expression ofβ-cat, vascular endothelial cadherin (VE-cad), Twist-1 and E-cadherin (E-cad) in TPC-1 cells were detected by Western blotting.Results The expression level of miR-9-3p in TPC-1 cells in the inhibitory group was 0.21±0.10, significantly lower than that in the blank control group (1.02±0.16) and the empty transfection group (0.98±0.15). MTT results showed that the proliferative rates of TPC-1 cells at 24, 48, 72 and 96 h in the inhibitory group were (95.68±4.34) %, (87.46±4.59) %, (78.11±4.63) %and (71.24±3.91) %, which were significantly lower than empty transfection group (P<0.05). Transwell invasion assay showed that the number of invasive TPC-1 cells in inhibitory group was 157±62, which was significantly lower than blank control group and empty transfection group (P<0.05). Scratch test showed that the healing rate of TPC-1 cells in the inhibitory group was (34.02±7.11) %, significantly lower than that in the blank control group and the empty transfection group (P<0.05). The nuclear transfer ofβ-cat in TPC-1 cells was significantly less in the inhibitory group than in the blank control group and the empty transfection group.The expression levels ofβ-cat, VE-cad and Twist-1 protein in TPC-1 cells in the inhibitory group were 1.15±0.29, 0.23±0.08and 0.65±0.20, lower than blank control group and empty transfection group (P<0.05);while the protein level of E-cad in TPC-1cells in inhibitory group was 0.85±0.17, higher than blank control group and blank transfection group (P<0.05). Conclusion Down-regulation of miR-9-3p can inhibit the activation of Wnt/β-cat signaling pathway and reverse the epithelial-mesenchymal transition of TPC-1 cells, thereby reducing the invasion and migration of PTC.
