Expression of EphA7 in oral squamous cell carcinoma and its correlation with cancer stem cell

Abstract:Objective To investigate the expression of EphA7 in oral squamous cell carcinoma (OSCC),and the correlation of its expression with clinicopathological features and cancer stem cells (CSC). Methods Immunohistochemistry was used to analyze the expression of EphA7 in 54 cases of OSCC tissues (40 cases of primary OSCC tissues,7 cases of recurrent OSCC tissues after radical operation combined with radiotherapy and 7 cases of recurrent OSCC tissues after radical operation combined with chemotherapy) and 14 paracancerous tissues. Subsequently,the EphA7 gene in OSCC cell line CAL27 was silenced by siRNA in vitro to detect the ability of sphere formation. The expression of stem cell markers was detected by Western blotting. Results The overexpression rates of EphA7 in OSCC tissues and adjacent normal tissues were 58.9% (33/56) and 14.3% (2/14) with significant difference (P<0.05). The overexpression of EphA7 in recurring OSCC after radiotherapy and chemotherapeutic tissues were both 85.7% (6/7), which were higher than 50% (20/40) of primary OSCC with significant difference (P<0.05). In CAL27 cells, knock-out of EphA7 reduced sphere formation rate from 57.0%. to 21.3%., and down-regulated expressions of stem cell markers (ALDH1,BMI1 and OCT4). Conclusion EphA7 may play an important role in the stemness maintenance of OSCC and may present a potential therapeutic target for OSCC.
