Effects of rhizoma polypodiodis nipponicae extracts on the proliferation and apoptosis of cervical cancer cells

Abstract:Objective To investigate the effects of rhizoma polypodiodis nipponicae extracts (RPNE) on the proliferation and apoptosis of cervical cancer cells in vitro and in vivo. Methods The cervical cancer Hela cell lines were divided into the control group and the RPNE treatment group after the logarithmic growth. After treatment with 2,4,and 8 μg/ml RPNE for 48 h or 8 μg/ml RPNE for 12,24, and 48 h,cell morphology was observed by phase contrast inverted microscope and cell proliferative activity was detected by MTS cell activity method. Flow cytometry using Annexin V/PI double staining method was employed to detect the apoptotic rates of2, 4 and 8 μg/ml RPNE after 48 h. Xenograft tumor growth inhibitive assay was performed to measure the anti-cancer effects of RPNE in vivo. Results It was observed under the phase contrast microscope that the growth inhibition and apoptosis of Hela cell lines were significantly induced by RPNE treatment over 4 g/ml at 48 h. The results of MTS detection of cell relative activity showed that RPNE treatment could inhibit cell activity in a concentration-and time-dependent manner (P<0.05). Annxin V-FITC/PI double staining flow cytometry analysis showed that compared with the 0 μg/ml,apoptotic rates of 2-8 μg/ml RPNE increased in a concentration-dependent manner (P<0.05). The volume of tumor in the control group was accelerated at 8 days after inoculation, while the RPNE group began to decrease. After treatment for 10 days, the tumor volume of RPNE group was lower than that of the control group (P<0.05),and tumor weight of RPNE group was lower than that of the control group (P<0.05). Conclusion RPNE can induce apoptosis and proliferation inhibition of cervical cancer cells in vitro,and can effectively induce the growth inhibition of Hela originated xenograft in nude mice, having potential in the treatment of cervical cancer.
