The role of NRP-1 mediated VEGF-VEGFR2-independent pathway in tumorigenesis

Abstract:Neuropilin?1 ( NRP?1) , an important member of NRP family, is often expressed in endothelial cells and some cancer cells. Recent studies suggest that NRP?1 promotes tumor development mainly via VEGF?VEGFR2 pathway. In addition, NRP?1 also binds with other growth factors such as platelet derived growth factor ( PDGF) and its receptors. Moreover, recent studies show that NRP?1 promotes angiogenesis via NRP?1?ABL pathway, but independent of VEGF?VEGFR2. Downstream of NRP?1?ABL, PDGF sig?naling, is related to RAD51 protein, the key enzyme that catalyzes homologous recombination of DNA repair. Overexpression of RAD51 induces cancer cell resistance to radiotherapy and chemotherapy. Inhibition of NRP?1 may overcome the limitations of individual inhibi?tion of VEGF?VEGFR2 pathway in cancer therapy, and provide new ideas for cancer treatment. Therefore, we review the role of NRP?1 in tumorigenesis and development independent of VEGF?VEGFR2 pathway.
