The correlation of HER-2 gene amplification and Ki-67 protein expression in gastric cancer

Abstract:Objective To investigate the expression and relationship of HER?2 and Ki?67 in gastric cancer, and analyze their correlation with clinicopathological parameters. Methods FISH and immunohistochemical staining were used to detect the HER?2 gene amplification and Ki?67 protein expression in 156 gastric cancer tissues. The association between HER?2, Ki?67 expressions and clini?copathological features was analyzed. The interrelationship of HER?2 and Ki?67 expression in gastric cancer tissues was explored. Re?sults The amplification rate of HER?2 gene was 11?5%( 18/156) . The amplification rate of HER?2 gene was significantly related with depth of invasion and cancer embolus(P<0?05). The positive expression rate of Ki?67 in gastric cancer tissues was 73?72%(115/156) , which was higher than 2?56%( 4/156) of para?carcinoma tissues with significant differences( P<0?05) . The expression of Ki?67 was significantly related with histological type and depth of invasion. HER?2 amplification and Ki?67 expression was positively correlated(r=0?162, P<0?05). Conclusion HER?2 and Ki?67 expression may be involved in the tumor progression of gastric cancer. The combination detection of HER?2 amplification and Ki?67 expression may be used to evaluate the patients?prognosis and de?velop an individualized therapy.
