Expressions of circRNA0084043,miR-31 and KLF8 in cancer tissues and their association with prognosis of patients with cutaneous malignant melanoma

Abstract：Objective To investigate the expressions of circRNA0084043,miR-31 and KLF8 in cancer tissues,and their relationship with prognosis of patients with cutaneous malignant melanoma(CMM).Methods A total of 80 patients with CMM in the hospital and 40 patients with pigmented nevus were enrolled between September 2022 and August 2024 as CMM group and control group,respectively.The expression levels of circRNA0084043,miR-31 and KLF8 were compared between the CMM group and the control group,which were analyzed in CMM patients with different basic characteristics.The correlation among the expressions of circRNA0084043,miR-31 and KLF8 in cancer tissues of CMM patients was analyzed by Pearson correlation analysis.After 36 months of follow-up,the relationship of circRNA0084043,miR-31,KLF8 expressions with prognosis and survival was analyzed by Kaplan-Meier method.Results The expressions of circRNA0084043 and KLF8 in the CMM group were higher than those in the control group,while miR-31 was lower than that in the control group(P<0.01).There were significant differences in expressions of circRNA0084043,miR-31 and KLF8 among CMM patients with different tumor grade,tumor sites,differentiation degree and lymph node metastasis(P<0.01).Pearson correlation analysis showed that circRNA0084043 expression in cancer tissues of CMM patients was negatively correlated with miR-31 expression(r=-0.454,P<0.01),but positively correlated with KLF8 expression(r=0.427,P<0.01).The expression of miR-31 was negatively correlated with KLF8 expression(r=-0.331,P<0.01).The 36-month overall survival rate and median survival time were lower or shorter in patients with high expressions of circRNA0084043 and KLF8 than in those with low expressions(P<0.05),which however,were higher or longer in patients with high expression of miR-31 than in those with low expression(P<0.01).Conclusion The expressions of circRNA0084043 and KLF8 are increased,while miR-31 is decreased in cancer tissue of CMM patients,and expressions of all the three indicators are correlated with prognosis.
