Research progress on the mechanism of Nrf2/HO-1 signaling pathway in drug-induced liver injury

Abstract:Drug-induced liver injury(DILI)refers to liver damage caused by medications or their metabolites and is one of the significant types of liver diseases in clinical practice.With the continuous expansion of clinical medication varieties,the incidence of DILI has shown a significant upward trend,seriously threatening people's health.In recent years,it is found that nuclear factor erythroid 2-related factor-2(Nrf2)/heme oxygenase-1(HO-1)signaling pathway plays an important role in the development of DILI and is expected to become a novel target for the treatment of DILI.The Nrf2/HO-1 signaling pathway is a multi-target and multi-mechanism signaling pathway that serves as the core regulatory system for cellular antioxidant stress response.It can enhance the antioxidant stress response through mechanisms such as upregulating the expression of antioxidant enzymes,inhibiting inflammatory factors,regulating apoptosis and iron metabolism,and controlling autophagy,and it can dynamically regulate the oxidative environment in the body,and effectively reduce the degree of liver tissue damage.
