Expression and Significance of mRNA and Protein Related to the JAK2/STAT3 Signaling Pathway in Tissues of Patients with EGFR Mutation-positive Non-small Cell Lung Cancer

Abstract:Objective To investigate the expression and significance of mRNA and protein related to the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway in the tissues of patients with epidermal growth factor receptor(EGFR)mutation-positive non-small cell lung cancer(NSCLC).Methods Ninety patients with EGFR mutation-positive NSCLC who visited the hospital from January 2020 to July 2023 were selected as the positive group.Gender and age were used as matching factors,and 90 patients with EGFR mutation-negative NSCLC during the same period were selected as the negative group according to the 1︰1 matching principle.The baseline characteristics of the two groups,and the mRNA and protein expressions related to the tissue JAK2/STAT3 signaling pathway were statistically analyzed.Spearman rank correlation analysis was used to analyze the correlations among the indicators.The efficacy of JAK2/STAT3 signaling pathway-related mRNA and protein in diagnosing EGFR mutation-positive NSCLC was analyzed by using the receiver operating characteristic(ROC)curve,and the impact of JAK2/STAT3 signaling pathway-related mRNA and protein expression on the 1-year prognosis of EGFR mutation-positive NSCLC patients was analyzed by using the K-M survival curve.Results The mRNA and protein of JAK2 and STAT3 in the positive group were higher than those in the negative group(P<0.01).The mRNA and protein of JAK2 and STAT3 in the tissues of EGFR mutation-positive NSCLC patients were positively correlated with histological type,lymph node metastasis,tumor diameter,positive Ki-67,CEA,SCCAg,and CYFRA21-1(P<0.05,P<0.01).The ROC curve showed that the areas under the curve of JAK2 mRNA+JAK2 protein+STAT3 mRNA+STAT3 and CEA+SCCAg+CYFRA21-1 for diagnosing EGFR mutation-positive NSCLC patients were 0.934 and 0.921,respectively,and there was no significant difference(P>0.05).The 1-year survival rate of patients with high values of JAK2 mRNA and protein and STAT3 mRNA and protein was lower than that of patients with low values(P<0.01).Conclusion JAK2 and STAT3 mRNA and protein are highly expressed in the tissues of EGFR mutation-positive NSCLC patients,and are related to tumor diameter,histological type,lymph node metastasis,positive Ki-67,CEA,SCCAg,CYFRA21-1 and prognosis.The combined detection has a high diagnostic efficacy and is helpful for adjusting the treatment plan,and controlling the progression of the disease.
