circVANGL1/miR-134-5p/CDCA3 Axis Regulates Proliferation,Migration and Invasion of Breast Cancer Stem Cells

Abstract:Objective To explore the mechanism of circVANGL1/miR-134-5p/cell division cycle associated 3(CDCA3)axis regulating proliferation,migration and invasion of breast cancer stem cells.Methods Breast cancer tissues and corresponding adjacent tissues of patients treated for breast cancer from February 2021 to November 2023 were collected.Breast cancer MCF-7 cell stem cell subpopulation were cultured in vitro.According to different treatment methods,they were divided into Con group,si-NC group,si-circVANGL1 group,pcDNA group,pcDNA-CircvangL1 group,miR-NC group,miR-134-5p group,anti-miR-NC group,anti-miR-134-5p group and si-circVANGL1+anti-miR-134-5p group,and si-circVANGL1+pcDNA-CDCA3 group.qRT-PCR was used to detect the expression levels of circVANGL1,miR-134-5p and CDCA3 mRNA in breast cancer tissues,adjacent tissues and breast cancer stem cells of each group.The expression levels of CDCA3,matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)were detected by Western blot.Dual luciferase repor-ting experiments verified the targeting relationship between circVANGL1 and miR-134-5p and between miR-134-5p and CDCA3.The proliferative activity of breast cancer stem cells was detected by CCK-8 assay,the cell migration ability was de-tected by scratch assay,the cell clonogenesis ability was detected by clonogenesis assay,and the cell invasion ability was de-tected by Transwell chamber assay.Results Compared with adjacent tissues,the expression levels of circVANGL1,CDCA3 mRNA and CDCA3 protein in breast cancer tissues were higher,while the expression level of miR-134-5p was lower(P<0.05).circVANGL1 could target and regulate the expression of miR-134-5p,and miR-134-5p could target and regulate the expression of CDCA3 protein.After interfering the expression of circVANGL1,the proliferation activity,scratch healing rate,the number of cell clones formed,the number of invasive cells and the expression levels of MMP-2 and MMP-9 protein of breast cancer stem cells were significantly decreased(P<0.05).Down-regulation of miR-134-5p expression or overexpres-sion of CDCA3 could partially reverse the effect of interfering circVANGL1 on proliferation,migration and invasion of breast cancer stem cells(P<0.05).Conclusion CircVANGL1 may regulate the proliferation,migration and invasion of breast cancer stem cells through the miR-134-5p/CDCA3 axis.
