miR-29 Affects the Biological Behavior of Cervical Cancer Cells by Regulating CDK6

Abstract:Objective To investigate the mechanism of miR-29 regulating the biological behavior of cervical cancer cells by targeting CDK6. Methods Twenty patients with cervical cancer who received surgery but without any treatment before surgery were recruited in this study, to collect cancerous tissues and adjacent normal tissues. When the cells were grown to approximately 80% confluency in cell culture, they were passaged by digestion method and cryopreserved. The expression of miR-29 in cervical cancer tissues and adjacent normal tissues was detected by qPCR. The effect of miR-29 on the proliferation of cervical cancer cells was detected by MTT assay. In addition, the effect of miR-29 on the aggregation function of cervical cancer cells was assayed by Colony formation assay. Dual lu-ciferase assay validated the effect of miR-29 on CDK6 transcriptional activity. Results The expression of miR-29 in cervical cancer tissues was significantly lower than that in normal tissues [(1. 23 ± 0. 45) vs (1. 91 ± 0. 35), P<0. 001] . The results of MTT assay showed that miR-29 significantly inhibited the proliferation of Hela and Siha cells after transfected with LV-miR-29 or LV-miR-Ctrl. The results of Colony formation assay showed miR-29 significantly inhibited the aggregation ability of Hela and Siha cells. Moreover, dual luciferase results indicated a direct binding site between miR-29 and CDK6, and miR-29 regulated the transcriptional activity of CDK6. Conclusion miR-29 can inhibit the proliferation and aggregation ability of cervical cancer cells by targeting CDK6, which may serve as a novel target marker for cervical cancer.
