Diagnostic Value of Thinprep Cytology Test Combined with Detection of BRAF Gene Mutation in the Diagnosis of Thyroid Nodules

Abstract:Objective To investigate the diagnostic value of thinprep cytology test (TCT) combined with detection of serine/threonine-protein kinase (BRAF) gene mutation in the diagnosis of thyroid nodules.Methods We enrolled a total of 108 patients with thyroid nodules confirmed by surgery and pathological examination in our hospital from April 2014 to April 2017.The conventional cell smear (CS),TCT and detection of BRAF gene mutation were performed.The sensitivity,specificity and accuracy of different examination methods were compared in terms of the diagnosis of benign and malignant thyroid nodules.Results There were 25 cases of malignant tumor confirmed by postoperative pathology and 83 cases of benign lesions.Pathological examination results served as the golden standard.The sensitivities of CS,TCT,BRAF gene mutation detection,and TCT combined with BRAF gene mutation detection in the diagnosis of malignant thyroid tumors were 76.00%,92.00%,64.00% and 96.00% respectively;the specificities and accuracy rates were 78.31%,96.39%,100.00%,100.00% and 77.78%,95.37%,91.67%,99.07% respectively.With respect to the diagnosis of thyroid malignant tumors,the sensitivity and accuracy of TCT combined with BRAF gene mutation detection were significantly higher than those of CS or BRAF gene mutation detection,with the specificity higher than that of CS (P < 0.05).Conclusion TCT combined with BRAF gene mutation detection can improve the diagnostic rate of thyroid nodules.
