Experimental Study of Effect of Aliskiren in Treatment of Renal Damage Induced by Ureteral Obstruction

Abstract:Objective To investigate effect of Aliskiren in treatment of renal damage induced by ureteral obstruction. Methods Animal models of complete left ureteral obstruction were established in New Zealand rabbits, and then 16 success-ful animal models were randomly divided into experimental group (n=8) and control group (n=8). Experimental group was treated with 25 mg/kg/d Aliskiren orally, while control group was followed up only. Glomerular filtration rates ( GFR) and re-nal blood flow resistance index (RI) of left kidney before and in the 2nd, 4th and 8th weeks after the experiment, and pathologi-cal results of left kidney 8th weeks after the experiment were observed and compared respectively in two groups. Results There were no significant differences in GFR and RI of left kidney before experiment in two groups (P>0. 05). In the 2nd, 4th and 8th weeks after experiment, the GFR values were significantly lower, while RI levels were significantly higher than those before experiment than those before experiment in two groups (P<0. 05); in experimental group, GFR value of left kidney was significantly higher, while RI level of left kidney was significantly lower than those in control group, and there were signif-icant differences in GFR values of left kidney in the 2 nd , 4 th and 8 th weeks after experiment and RI level in 8 th weeks after ex-periment between the two groups (P<0. 05). Degrees of fibrosis and inflammatory reaction of left renal in 8th weeks after ex-periment in experimental group were significantly slighter than those in control group. Conclusion Aliskiren may improve blood flow perfusion of renal damage induced by ureteral obstruction GFR and decrease degrees of fibrosis and inflammatory re-action in New Zealand rabbits.
