Metabolism of TDP-43 in Human Embryonic Kidney Cells and Effect of Its Production on Cell Viability

Abstract:Objective To investigate the metabolic processes of TDP-43 in HEK293 ( human embryonic kidney cells) and effect of its production on cell viability. Methods The HEK293 was affected with different concentrations of Stau-rosporine (STS) to detect the protein level of TDP-43, and then metabolic processes were explored, combined with different blocking agents of protein degradation pathway, and the effect on cell viability was also observed by transfection ectogenesis TDP-43. Results STS had limited effect on protein level of TDP-43 in HEK293, but it could induce 35 kD fragment in high-er concentration (5 μM and 10 μM). The 35 kD fragment was degraded through ubiquitin proteasome system, and overex-pression of 35 kD fragment could obviously decrease cell viability. Conclusion Overexpression of TDP-43 and its 35 kD frag-ment can inhibit cell viability in HEK293, which is probably the cause of TDP-43 proteinopathies.
