Effect of ethanol extract of propolis on m6A methylation in colon cancer HCT116 cells

Abstract:This study investigates the inhibitory effects and underlying mechanisms of ethanol extract of Chinese propolis(EECP)on the proliferation of human colorectal cancer cells(HCT116)through modula-tion of N6-methyladenosine(m6A)RNA methylation.HCT116 cells were treated with varying concentra-tions of EECP(25,50,and 100 μg/mL).Cell proliferation was assessed using the CCK-8 assay,while the global m6A modification levels of total RNA were measured by dot blot analysis.The mRNA and protein expression levels of Methyltransferase-like 3(METTL3),Methyltransferase-like 14(METTL14),Wilms Tumour 1-Associating Protein(WTAP)and Fat Mass and Obesity-associated Protein(FTO)were evalua-ted using real-time quantitative PCR(RT-qPCR)and Western blot,respectively.The results demonstra-ted that increasing concentrations of EECP significantly reduced the total RNA m6A levels in HCT116 cells(p<0.05).Additionally,EECP treatment led to decreased expression levels of FTO,WTAP and METTL3,while METTL14 expression was significantly upregulated(p<0.05).In conclusion,EECP induces alterations in m6A methylation and related enzyme expression in HCT116 cells,suggesting that EECP may exert anti-colorectal cancer effects by regulating m6A methylation.
