Advances in miscibility prediction and performance optimization of amorphous solid dispersions based on polymer selection

Abstract:Amorphous solid dispersion(ASD)have attracted considerable attention in drug delivery as a key strategy for enhancing the solubility and bioavailability of poorly soluble drugs.Polymer selection plays a pivotal role in ASD systems,directly influencing drug-polymer miscibility,stability,and overall formulation performance.Consequently,accurately predicting drug-polymer miscibility and selecting suit-able polymers based on specific performance requirements are of significant research interest in ASD opti-mization.This review highlights the critical role of polymer selection in ASDs,systematically summarizes both predictive and experimental methods for assessing drug-polymer miscibility,and comprehensively ex-amines various performance optimization strategies,including,recrystallization inhibition,dissolution en-hancement and stability improvement.By providing theoretical insights into the rational design of ASDs,this review aims to facilitate their broader application in the delivery of poorly soluble drugs.
