Construction of magnetic-assisted aptamer sensor based on screen-printed electrode for electrochemical detection of H1N1 in wastewater

Abstract:The H1N1 influenza virus,as a highly contagious pathogen,can exacerbate the risk of spread through water environment media,posing a severe threat to public health security.In wastewater testing,traditional detection methods for H1N1 face challenges such as insufficient sensitivity due to complex ma-trices,cumbersome and time-consuming procedures,and difficulty in achieving rapid on-site screening.Electrochemical aptamer sensors,with their high sensitivity,fast response,and excellent interference re-sistance,are particularly suitable for detecting samples from complex environments.This study aims to develop a miniaturized sensor for on-site,real-time detection of H1N1 influenza virus in wastewater.Two aptamers were modified onto Fe3O4-NH2@Au magnetic nanoparticles and Au@Pt nanoenzymes surfaces,respectively,to capture the H1N1-HA protein and form a composite structure of capture component-tar-get-identification component.This structure was then fixed onto a screen-printed electrode via an external magnetic field,catalyzing the H2O2/hydroquinone reaction to detect the H1N1-HA protein.In actual wastewater samples,the sensor demonstrated good recovery rates for the H1N1-HA protein,and the sen-sor based on screen-printed electrodes is more convenient for on-site real-time detection.
