Box-Behnken Design Optimization of Crude Polysaccharide from Dregs of Cichorium Glandulosum Enzyme Extraction Process

Abstract:In order to achieve efficient extraction and rational utilization of the active ingredient,crude polysaccharides from the residue of Xinjiang's characteristic plant medicine, Cichorium glandulosum,the response surface methodology was used to optimize the extraction process conditions of crude polysaccharides from Cichorium glandulosum. The content of fructose and glucose in the extracted crude polysaccharides was identified by High Performance Liquid Chromatography-Refractive Index Detector(HPLC-RID),and their in vitro antioxidant activity was finally investigated. Using the medicinal residue of Cichorium glan-dulosum root after alcohol extraction as the raw material,the yield of crude polysaccharides from Cichorium glandulosum root was used as the evaluation index, and the extraction temperature,extraction time, solid-liquid ratio, and enzyme amount were the main influencing factors. Based on single factor experi-menu,the response surface methodology was used to optimize the extraction process of crude polysaccha-rides from Cichorium glandulosum root. The content of fructose and glucose in the crude polysaccharides of Cichorium intyhus was identified by HPLC-RID according to the national standard method. The in vitro antioxidant activity of the crude polysaccharides of Cichorium glandulosum was examined by 1,1-diphenyl-2-picryl-hydrazyl radical (1,1-diphenyl-2-picryl-hydrazyl radical,DPPH) and 3-ethylhenzothiazoline-6-sul-fonic acid (3-ethylhenzothiazoline-6-sulfonic acid) (ABTS).The optimal solution for optimizing the extraction of crude polysaccharides from Cichorium glandulosum seed residue using the response interview method is an extraction time of 2. 0 hours,enzyme content of 2. 0%,solid-liquid ratio of 1,40(g?mL-1),extraction temperature of 51℃.The yield of polysaccharides extracted from Cichorium glandulo-sum seed residue under these conditions is(20. 64±0. 34)%.HPLC-RID method can achieve efficient separation and identification under the conditions of acetonitrile: water(75:25,V:V) as the mobile phase,flow rate of 1. 0 mL?min-1,and column temperature of 35℃.In vitro antioxidant tests show that crude polysaccharides from Cichorium glandulosum seed have good scavenging effects on DPPH radicals and ABTS radicals. This study obtained the optimal extraction process of crude polysaccharides from Ci-thorium glandulosum by enzymatic method,and identified two monosaccharide components,fructose and glucose,in the crude polysaccharides of Cichorium glandulosum.It also demonstrated that the crude poly-saccharides of Cichorium glandulosum have good in vitro antioxidant activity.
