An animal experimental study on the repair of intervertebral disc annulus fibrosus with oxidized regenerated cellulose and TGF-β3

Abstract:Objective To explore the therapeutic effect of oxidized regenerated cellulose combined with transforming growth factor(TGF-β3)in repairing intervertebral disc annulus fibrosus,and provide experimental evidence for clinical application.Methods Forty rabbits were randomly divided into four groups.The L2-3,L3-4 and L4-5 intervertebral discs were selected to expose the intervertebral disc annulus fibrosus.The blank control group,model group,suture group and experimental group were established,with ten rabbits in each group.In the control group,only the corresponding intervertebral discs were exposed.In the other three groups,the corresponding annulus fibrosus was incised.The model group was not given any treatment.The suture group was treated with simple suture and the annulus fibrosus incision was repaired.The experimental group was treated with oxidized regenerated cellulose combined with TGF-β3 for suture of the annulus fibrosus incision.Twelve weeks after operation,five rabbits in each group were randomly selected for biomechanical detection,and the nucleus pulposus leakage pressure was recorded.The rest of the rabbits in each group were detected by MRI before the corresponding intervertebral disc tissues were obtained for pathological section staining and observation.Results The nucleus pulposus leakage pressure in the experimental group was higher than that in the model group or suture group,but lower than that in the control group(P<0.05).There was no statistically significant difference between the model group and the suture group(P>0.05).Twelve weeks after operation,the T2 value in the experimental group was higher than that in the model group or the suture group,and the intervertebral height loss was milder than that in the model group or the suture group.However,the T2value in the experimental group was lower than that in the model group and intervertebral height loss was more severe than that in the control group(P<0.05),but there was no statistically significant difference between the model group and the suture group(P>0.05).Pathological staining showed that the fusion of the original annulus fibrosus and implanted material was good in the experimental group,and the level of the annulus fibrosus was slightly worse than that in the control group.In the model group and the suture group,there was continuous damage to the annulus fibrosus,filled by scar tissues.The content of type Ⅱcollagen was decreased.Conclusion The oxidized regenerated cellulose combined with TGF-β3 plays an active role in repairing intervertebral disc annulus fibrosus defects.
