Long non-coding RNA Florpar induces apoptosis in cerebrovascular smooth muscle cells in simulated microgravity rats

Abstract:Objective To investigate the effects of long non-coding RNA Florpar on cerebrovascular remodeling of simulated microgravity rats.Methods A high throughput microarray assay was performed to screen the changes of LncRNA expressions in cerebral arteries of 4-week hindlimb unweighting rats.Aortic smooth muscle cells A7r5 were transfected with LncRNA-Florpar plasmid or siRNA respectively and the apoptotic rate was examined by Hoechst+PI staining.The expressions of apoptosis-related proteins(Bcl-2 and Bax)were also assessed by Western blotting.Results Compared with control rats,the expression of LncRNA-Florpar was significantly increased in cerebral arteries of rats that had undergone 4-week simulated microgravity.Furthermore,upregulation of LncRNA-Florpar significantly increased the apoptotic rate and the ratio of Bax/Bcl-2 in A7r5 cells.In contrast,downregulation of LncRNA-Florpar significantly decreased the apoptotic rate and the ratio of Bax/Bcl-2 in A7r5 cells.Conclusion We have found for the first time that the expression of LncRNA-Florpar can be significantly increased in cerebral arteries of simulated microgravity rats.Furthermore,LncRNA-Florpar can induce apoptosis in vascular smooth muscle cells.The present study indicates that LncRNA-Florpar can sense the change of mechanical signals in a microgravity environment and then participate in cerebral arterial remodeling by regulating the apoptosis of VSMCs.
