Effects of miR-218 on proliferation and invasion of leukemia cells through SNX4

Abstract:Objective To investigate the effect of miR-218 on the proliferation and invasion of leukemia cells and the mechanism. Methods The expression of miR-218 in different acute lymphoblastic leukemia cell lines was detected with qPCR. Double luciferase reporter gene was used to detect the mutual regulation between miR-218 and SNX4. The effect of miR-218 expression on the proliferation of E6-1 cells was detected with cell proliferation assay. Transwell invasion assay was used to detect the effect of miR-218 expression on E6-1 cell invasion. Results Compared with other cell lines, the expression of miR-218 in E6-1 cell line was significantly decreased. miR-218 could specifically bind to the 3'UTR of SNX4 and regulate the expression activity of SNX4. Overexpression of miR-218 could inhibit the proliferation and invasion of E6-1 cells. Conclusion The expression level of miR-218 in acute lymphoblastic leukemia is significantly decreased, which can inhibit the proliferation and invasion of E6-1 cells by regulating the expression of SNX4.
